THE COURSE AIMS TO PROVIDE THE KNOWLEDGE OF THE MAIN METHODS FOR THE STUDY OF THE MACROMOLECULES OF BIOLOGICAL INTEREST (PROTEINS, NUCLEIC ACIDS), WITH PARTICULAR REFERENCE TO PRODUCTION STRATEGIES, PURIFICATION AND CHARACTERIZATION OF THESE MACROMOLECULES. THE MAIN KNOWLEDGE ACQUIRED BY THE STUDENT WILL CONCERN: -THE CHEMICO-PHYSICAL FEATURES AND THE BIOLOGICAL ROLE OF BIOLOGICAL MACROMOLECULES AND HOW THESE FEATURES CAN BE USED FOR THEIR PURIFICATION AND CHARACTERIZATION; - THE DIFFERENT METHODS OF PURIFICATION OF PROTEINS AND NUCLEIC ACIDS: METHODS OF HOMOGENIZATION OF CELLS AND TISSUES, DIFFERENTIAL PRECIPITATION, CENTRIFUGATION AND DIALYSIS, CHROMATOGRAPHY; -THE DIFFERENT ANALYTICAL METHODS RELATED TO PROTEINS AND NUCLEIC ACIDS: ELECTROPHORESIS, SPECTROSCOPIC METHODS, SEQUENCING METHODS , IMMUNOCHEMICAL METHODS, TECNIQUES OF BLOTTING. -THE MAIN METHODS OF THE RECOMBINAT DNA. THE MAIN SKILLS DEVELOPED BY THE STUDENT WILL BE: -TO KNOW AND EXPOSE THE CHARACTERISTICS OF THE VARIOUS TECHNIQUES STUDIED -TO CRITICALLY EVALUATE PROTEIN PRODUCTION AND PURIFICATION PROTOCOLS; -TO EVALUATE THE STRUCTURAL AND FUNCTIONAL CHARACTERISTICS OF PROTEINS AND NUCLEIC ACIDS ON THE BASIS OF RESULTS OBTAINED WITH DIFFERENT TECHNIQUES; -TO PERFORM SIMPLE PROTOCOLS FOR PURIFICATION AND CHARACTERIZATION OF PROTEINS AND NUCLEIC ACIDS. |

THE TOPICS PRESENTED IN THE CLASSROOM LESSONS ARE: - STRUCTURAL CHARACTERISTICS OF PROTEINS AND NUCLEIC ACIDS (ABOUT 2 H FRONTAL LESSONS) - CELLULAR CULTURES; VIABILITY ASSAYS (ABOUT 3 H FRONTAL LESSONS) - METHODS FOR PROTEIN AND DNA PURIFICATION: HOMOGENEIZATION OF CELLS AND TISSUES, DIFFERENTIAL PRECIPITATION METHODS, CENTRIFUGAL TECHNIQUES (ABOUT 5 H FRONTAL LESSONS) CHROMATOGRAPHIC TECHNIQUES (ABOUT 5 H FRONTAL LESSON) - ELECTROPHORETIC METHODS FOR PROTEINS AND NUCLEIC ACIDS(ABOUT 5 H FRONTAL LESSON) - SPECTROSCOPIC METHODS FOR THE STUDY OF PROTEINS AND NUCLEIC ACIDS (ABOUT 4 H FRONTAL LESSON) - TECHNIQUES OF SEQUENCING AND RADIOISOTOPES (ABOUT 5 H FRONTAL LESSON) –IMMUNOCHEMISTRY (WESTERN BLOT, IMMUNOPRECIPITATION, IMMUNOENZYMATC ASSAYS)(ABOUT 4 H FRONTAL LESSONS) -INTRODUCTION TO RECOMBINANT DNA TECHNOLOGY (HYBRIDIZATION, PCR, CLONING, RECOMBINANT PROTEINS) (ABOUT 7 H FRONTAL LESSONS). LABORATORY EXPERIENCES: PRACTICAL EXPERIENCES ARE FOCUSED ON PREPARATION OF PROTEIN EXTRACTS FROM CELL CULTURES, COLORIMETRIC ASSAYS, ELECTROPHORESIS, PCR (12 HOURS). |

THE TEACHING PROVIDES 52 HOURS OF TEACHING BETWEEN LESSONS, EXERCISES AND PRACTICAL LABORATORY. IN PARTICULAR, THERE ARE 40 HOURS OF CLASSROOM LESSONS (5 CFUS) AND 12 HOURS OF ASSISTED BIOCHEMISTRY LABORATORY EXERCISES (1 CFU). CLASSROOM LESSONS WILL BE HELD WITH THE HELP OF PROJECTIONS. |

ACHIEVING THE OBJECTIVES OF TEACHING IS CERTIFIED BY EXAMINATION. THE EXAMINATION PROVIDES AN ORAL TEST THAT CONSISTS OF AN INTERVIEW WITH QUESTIONS AND DISCUSSION ON THE THEORETICAL AND METHODOLOGICAL CONTENTS INDICATED IN THE TEACHING PROGRAM AND IS DESIGNED TO ASCERTAIN THE LEVEL OF KNOWLEDGE AND UNDERSTANDING ACQUIRED BY THE STUDENT AS WELL AS TO VERIFY THE ABILITY TO EXPOSURE BY USING THE APPROPRIATE TERMINOLOGY AND THE ABILITY TO AUTONOMOUSLY ORGANIZE THE EXHIBITION ON THE SAME SUBJECTS WITH THEORETICAL CONTENT. ALSO THE KNOWLEDGE OF THE DIFFERENT TOOLS AND APPLICATIONS LEARNT DURING THE EXERCISES WILL BE EVALUATED BY THE ORAL TEST. |
